研究概要	上皮細胞や内皮細胞とマトリックスとの間に存在する薄い基底膜を大量に産生し、人工皮膚、人工血管等の人工臓器において細胞とマトリックスとのあいだに挿入することによってより強固で、生体内と同じ構成の人工臓器を作製することを究極の目的としている。この一連の研究のなかで、私達が単離したα(IV)コラーゲン鎖をコードするcDNAを基にα(IV)コラーゲン分子を大量に作製することにある。本年度は6種のヒトα(IV)鎖をコードする各全長cDNAコンストラクトを作製することを目標に以下の実験を行った。六種類のIV型コラーゲン遺伝子の組織特異的な発現を種々の系で、分子が少なくとも三つの構成:α1/α1/α2、α3/α4/α5、α5/α5/α6があることを突き止めることができた。これは、ペプチドを抗原とするα鎖に特異的モノクローン抗体を用いた免疫組織化学染色、その二重染色、各α鎖の一次構造の比較からのコラーゲン遺伝子系統図から由来するコラーゲン分子の仮設に由来するものである。この仮設は生化学的もしくは分子生物学的方法によって証明する必要がある。このためにも各α(IV)コラーゲン鎖をコードする全長cDNAコンストラクトの作成が必須となる。全長α6(IV)鎖をコードするcDNAコンストラクトを作成することができた。これを無細胞系で翻訳すると、予想きれるサイズのタンパクのバンドが検出された。これが、α6(IV)鎖を認識する特異抗体と反応するかどうかを検定するための実験を現在行っている。α5(IV)鎖をコードするcDNAコンストラクトの作成が現在進行中である。
