新規セリンリン脂質特異的ホスホリパーゼの病態生化学とその応用

研究概要	ホスファチジルセリン(PS)は、PKC、DGキナーゼなどの活性化因子であるとともに、アポトーシスや血小板の活性化に伴って細胞表面に露出し、細胞が貪食される際のマーカーや血液凝固反応の足場として機能する。また、PSのアシル基が1つ加水分解されて生じるLysoPSは、ラット腹腔マスト細胞の活性化に必須であること、ヒト末梢T細胞の増殖を抑制することなどから、生理活性を持つ機能性リン脂質として位置づけ得る。我々はこれまでにPSの1位脂肪酸を特異的に加水分解する(PS-PLA1)の生化学的解析を行ってきた。今年度、(1)本酵素の発現が生体内の様々な部位で誘導されることを見出した。また、本酵素の生理的意義を明らかにするため、(2)ノックアウトマウスおよび過剰発現マウスを作製した。 (1)ラットにおけるリポ多糖(LPS)投与によるエンドトキシンショック、胸膜炎、腹膜炎、脳虚血・再灌流において、PS-PLA1の発現が著しく上昇することを見いだした。脳虚血・再灌流以外は、24時間以内に誘導が起こる早い反応であったが、脳虚血・再灌流の場合は、2週間以上にわたっておこるゆっくりとした反応であった。 (2)PS-PLA1の発現量が種々の刺激に応じて変動することを見出したが、この現象の意義や生体への影響を調べるツールとして、本酵素のノックアウトおよび過剰発現マウスを作製した。過剰発現マウスについては全身での発現レベルを上げることを目的として、b-アクチンプロモーター下にラットPS-PLA1翻訳領域を挿入したベクターを構築した。両者とも常法通りに作製し、ノックアウト1系統、過剰発現3系統を樹立した。いずれの系統のマウスも正常に発生し、外見上の異常は認められなかった。
