非IgE型刺激による肥満細胞活性化機構とその生物薬学的意義の解明

研究概要	肥満細胞をIgE非依存的に活性化する種々のアゴニスト(G蛋白型、サイトカイン類)の作用機序を明らかにするため、マウス骨髄由来未成熟肥満細胞(BMMC)を成熟型肥満細胞に(CTMC)様の細胞に分化誘導する系を確立した。具体的には、IL-3依存的に増殖するBMMCを別の肥満細胞増殖因子であるSCF存在下、繊維芽細胞と数日間共培養することにより、形態的にCTMC様の細胞染色像を示すと同時に、CTMCの機能的指標であるG蛋白共役型塩基性アゴニスト(非IgE型刺激)に対する応答性を獲得した均一の細胞集団を得た。この分化誘導過程は繊維芽細胞と肥満細胞の直接の接触により強く増強され、既知のサイトカインあるいは細胞接着因子以外の比較的不安定な繊維芽細胞由来因子が関与していることが明かとなった。また、非IgE型刺激に対する応答性が細胞膜上の糖蛋白の発現に依存する可能性を示唆する結果を得た。非IgE型刺激応答性の肥満細胞への分化誘導過程で発現が変化する遺伝子群を一括同定するため、cDNA subtraction法を用いて誘導型遺伝子群が高度に濃縮されたcDNA libraryを作製した。これを元に個々のクローンの配列決定を行い、現在までに約100個の既知または新規誘導型遺伝子を同定するに至っている。誘導型遺伝子群の約半数はマウス第14染色体上にコードされたセリンプロテアーゼ群で、中でもMMCP-4とGranzyme Bの誘導が際立っていた。これらプロテアーゼに次いで比較的高頻度で同定された遺伝子はNdr-1という名称でESTに登録されていた機能不明の細胞質蛋白であった。Ndr-1を強制発現した肥満細胞は脱顆粒反応が有意に亢進したことから、分泌応答の制御因子として機能している可能性を想像している。また、新規G蛋白共役型受容体様分子が同定された。現在、この分子が非IgE型刺激の受容体として機能している可能性を含め、その機能を解析中である。
