質量分析計を用いたHLA-DNAタイピングの自動化機械の開発

研究概要	1) 現在使用しているHLA-DRB1遺伝子増幅用のプライマーの中でも、比較的遺伝子型と分子量が検討容易であるDR4グループを用いて、PCR反応後の検体を質量分析計にて測定を行った。最初に、各対立遺伝子ごとに、塩基配列の違いより予想される分子量の算定を行ったところ、予想される分子量は171,667〜172,987と推定されたが、塩基配列の異なる対立遺伝子間でも、各塩基数が同一の場合は同一の分子量を示し、判別が困難なことが予想された。そこで一部の対立遺伝子については、塩基配列の違いを認識する制限酵素で切断してから用いるという方法を考案した。 2) 1)で予想された分子量をもとに、既知HLA-DR4対立遺伝子のホモ接合体B細胞株30種についてPCRでの増幅を行い、増幅産物のうち、一部については制限酵素Msp Iにて切断した後、脱塩カラムにて脱塩、精製後、滅菌水に溶解して質量分析器に注入し、分子量の測定を行った。結果、対立遺伝子の分子量そのものの測定は可能であった。しかしながら高速液体クロマトグラフィー分析の際には、精製後に残存するわずかな夾雑物の混入をも測定するために、コンピューターでの判定がやや困難な状況にあることから、増幅産物の精製法については、今後さらに高度な精製法を検討する必要があると考えられた。 3) 遺伝子型判定のためのコンピューターソフトのプログラミングについては、すべてのDRBl遺伝子について解析が可能なように、分子量の予測を行ったが、2)で得られた状況をもとにして、夾雑物の混入をも考慮して判定が可能な様に改良を行う予定である。
