高感度、高分解能MS/MS/MS分析用高エネルギー衝突活性化開裂装置の開発

研究概要	質量分析において高エネルギー衝突活性化開裂による生体分子の構造解析を効率よく行う方法を立するために、新たに衝突活性化開裂室(コリジョンセル)の設計、試作を行った。この際、高いイオン透過率と効率よい衝突活性化開裂を実現するにあたり、高電圧印加型コリジョンセル(耐圧10kV)とセル内で生成したプロダクトイオンを再加速するための加速レンズ(左右2枚からなる)を試作した。また、それらを組み込んだイオン源ブロックは、従来のものと互換性があるように計、試作した。合わせて、高電圧供給用密封端子、最大5kVまで印加可能な低リップル高圧電源、電場一磁場同時掃引用制御基盤を製作した。試作装置を既設の磁場型質量分析計に組み込み、0-5kVの範囲でコリジョンセルと加速レンズ(コリジョンセル電圧の50-90%)に電圧をかけ、標準試料を用いてイオン透過率について検討した。その結果、10kVで加速したイオンに対してコリジョンセル電圧3kVという条件で、プロダクトイオンのエネルギー損失、イオン透過率という点で最もよい結果が得られた。現在、この条件のもとで、実用質量範囲であるm/z100-3000において、既知ペプチド、糖、脂質などの生体分子の測定を行い、得られる構造情報と測定感度についてデーターを収集し、試作装置の評価と仕様を作成している。また、多価イオン(エレクトロスプレーイオン化法により生成)をプリカーサーとして測定した場合についても同様に検討を進めている。
