超音波による薬剤効果増強システムの開発と評価

研究概要	超音波の薬剤効果増強に関して、装置作成、超音波造影剤の利用、および経皮吸収の促進について検討した. 1. 薬剤効果増強は超音波の周波数に依存すると思われる.このため任意の周波数発生が可能な多周波超音波発生装置の作製に着手した. 2. 超音波照射により人工的微小気泡である超音波造影剤が壊れ、超音波の化学的効果や機械的効果を増強することが考えられ、既に超音波による血栓溶解への応用が検討されている.本研究では超音波造影剤類似物質としてPVC-AN(polyvinylidene chloride-acrylonitryl copolymer)を用いた.PVC-ANは酸素溶存水溶液において超音波による過酸化水素生成(・HO_2再結合による)を増強した.また、超音波によるKIからのI_2遊離と細胞溶解を促進した.照射時の溶存気体の種類に依存するが、PVC-ANは超音波誘発化学・生物学的効果を増強することが判明した.超音波造影剤を併用し、薬剤効果を増強できる可能性を示した. 3. 超音波による薬剤の経皮吸収促進作用に係わる因子である角質層への影響と皮膚電気抵抗について検討した.既に検討を進めてきたノルアドレナリンやヒスタミンに加えてアセチルコリン(ACh)の経皮吸収におよぼす影響も調べた.ゲル状にした薬剤をラット腹部に塗布後、超音波を照射した.照射ヘッドに恒温冷却水を循環することにより温度上昇を抑制した.経皮吸収促進作用は塗布される皮膚状態に依存し、角質層が除去されると促進効果は消失した.超音波照射中、皮膚電気抵抗は低下し、その程度は超音波強度に依存した.血圧変化を調べたところ、超音波照射によりAChの角質層通過時間が短縮すると思われる結果を得た.
