レトロウイルスライブラリーを利用した新しい発現クローニング法の開発とその応用

研究概要

研究代表者らが開発し、1995年に発表したレトロウイルスライブラリーを利用した発現クローニング法は、応用範囲が広く種々の実験に利用できる。今回のプロジェクトでは、1)GFPとのcDNAの融合ライブラリーを作成し、蛋白質の細胞内局在によって遺伝子を同定する方法(FL-REX)および 2)レトロウイルスcDNAライブラリーを利用したシグナルシークエンストラップ法(SST-REX)を開発して新規遺伝子をスクリーニングしている。FL-REX法では主に転写因子、SST-RE法ではサイトカインおよびサイトカインレセプターを探索している。現在までのところ、FL-REX法により胎児肝細胞からZnk Finger型の新規転写因子を1つ、SST-REX法により、T細胞から新規サイトカインレセプター、神経細胞から新規TNFレセプター、脂肪細胞から増殖因子様分子cDNAを同定し、現在解析中である。今後レセプターのリガンドのクローニングを行いレセプターの細胞内での作用を明らかにしていきたい。
