昆虫の脳内光受容器を中心とした長期現象としての光の受容処理システムの解析

研究概要

昆虫は生活圏を、海域を除く地球上のあらゆる環境に拡大し、繁栄している。昆虫にとって重要な環境情報のひとつである光は、短期的な現象を認識することに加え、日長の変化に対応した休眠・変態などの制御といった長期的な現象を認識する重要な刺激である。その長期現象を受容する器官は複眼の他に、おそらく脳内に別の器官あるいは光受容部位の存在が示唆されていたが、脳内光受容器官自身は不明であった。我々は本研究において甲虫ホタルを主に用いて、昆虫の成虫脳内に光受容器官が存在していることを明らかにした。
(1) 計12種の甲虫の脳を解剖し、脳内光受容器の存在と構造を観察したところ、内6種では視葉で、3種で脳のcaudal側で、残りの3種では観察されなかった。確認されたすべての光受容器で、マイクロビライがあり色素細胞で囲まれていた。
(2) 微小電極を用いて生理学的に光応答を記録すると、微弱光に反応する感度の高い応答性を示し、スペクトル応答極大は530nmであった。複眼のそれはそれぞれ560,380nmであることから明らかに起源の違う細胞であることが示唆された。
(3) 幼虫側単眼のスペクトル応答は530nmであり、幼虫時に左右一対あったものが生体脳ではcaudal側に一対となり、マイクロビライや周辺の色素細胞の構造など、幼虫側単眼と成虫脳内光受容器の形態がよく一致していた。
(4) 脳内光受容器の起源を検証するために、(1)幼虫側単眼除去個体の作成(2)幼虫側単眼にトレーサーをいれたものを作成し、継時的に追跡すると、幼虫側単眼が成虫脳光受容器に組み込まれることが確認された。
これらの結果から、幼虫側単眼の機能が、成虫脳内では変化していることが強く示唆され、動物は成長に従って感覚器官や神経系を変化させながら外界情報を新たに処理している可能性が示された。
