病原性機構の異なる2種の近縁な植物ウイルスにおける遺伝子間相互作用の研究

研究概要	植物RNAウイルスであるブロムモザイクウイルス(BMV)とササゲクロロティックモトルウイルス(CCMV)は、極めて近縁なウイルスであるにもかかわらず細胞間移行に外被タンパク質(CP)を必要とするか否かという点で、細胞間移行のメカニズムが大きく異なることが示唆されてきた。初年度の研究において、BMVとCCMVの細胞間移行においてCP要求性の違いを生じるウイルス遺伝子は移行タンパク質(MP)遺伝子であることが強く示唆された。本年度は、各ウイルスMPの機能解析およびMPとCPの間の相互作用の解析を試みた。これまで、BMVのMPには核酸結合活性があることが示されていたが、今年度はさらにCCMVのMPにも同様に核酸とくに一本鎖の核酸に対して配列非特異的な結合活性があることを明らかにした。この結果、BMV,CCMVともにRNA-MP複合体として細胞間を移行する可能性が示唆された。一方、BMVとCCMVのMP遺伝子の3'末端にGFP遺伝子を融合させ、プロトプラストに導入し発現させ、蛍光顕微鏡観察した結果、プロトプラスト表面に管状構造が観察された。この結果、両ウイルスとも原形質連絡中の管状構造中を粒子の形態で移行する可能性が示唆された。以上の結果、BMVとCCMVはそれぞれ細胞間移行におけるCPの要求性は異なるものの、いずれもRNA-MP複合体および粒子で移行する2つの可能性が示唆された。一方、BMVのMP遺伝子をグルタチオンS-トランスフェラーゼとの融合タンパク質として大腸菌で発現させ、純化し、BMV粒子から純化調整したCPとの結合をグルタチオンカラムを用いたpull-downアッセイにより調査した結果、BMVのMPとCPはin vitroで結合することが示された。今後、CCMVのMPについても同様の実験を行い、各ウイルスのMPのCP要求性と、両タンパク質間の結合活性の有無の相関関係を研究する際に今回確立したin vitro結合アッセイ系の利用が期待できる。
