センダイウイルスによる細胞融合反応の細胞側因子の解析

研究概要	我々はセンダイウイルスによる膜融合反応の分子機構を明らかにする目的で、ウイルス粒子によるヒト赤血球膜融合系で、膜骨格タンパク質の動態を解析した結果、融合に伴い膜骨格タンパク質バンド2.1(アンキリン)の特異的分解反応が認められた。一方F及びHN遺伝子発現によるHeLa等の培養細胞融合がμ-カルパイン阻害剤により抑制されることから、カルパインの関与が強く示唆されてきた。赤血球融合反応系でカルパインの活性化と膜骨格タンパク質の特異的分解反応を明確にすることにより、カルパインの新たな生理機能とこれまで明らかにされなかった融合反応の細胞側の制御機構を明らかにする。ヒト赤血球融合反応の時間経過及び融合効率とカルパインの活性化を中間型と活性化型カルパインの抗体を用いて再度詳細に解析した結果、融合溶血液でカルパインが活性化されていることが明らかになった。融合反応とアンキリンの限定分解はカルパイン阻害剤ZLLalおよびカルパインとプロテアソームの阻害剤ALLNalにより濃度依存的に阻害され、プロテアソーム阻害剤ラクタシスチンでは影響が認められなかった。また融合反応に際して、アンキリン以外の膜骨格タンパク質が特異的に分解されるか否かをスペクトリン、バンド3、アデューシンの特異抗体を用いてウエスタンブロットにより解析した結果、いずれの膜骨格タンパク質も融合特異的な分解は認められなかった。アンキリンの限定分解部位を同定することは、プロテアーゼの基質特異性を明らかにする上でも重要であり、限定分解物190kDaを精製し、切断部位のアミノ酸配列を試みたが、N末端がブロックされていた。おそらくカルパインの切断部位はC末端側に存在すると推定される。イオノフォア処理によるアンキリンの限定分解はカルシウムイオンの濃度に依存しており、カルパイン以外のカルシウム依存性の別の反応経路(例えばPKC等)を介していることも予想されるので、この可能性についても解析を進める予定である。
