真正細菌SRP RNAのドメイン構造と結合蛋白質、機能・構造相関

研究概要	グラム陽性細菌について、新たに5種類について構造解析を行った結果、調べた全てのSRP RNAには、枯草菌SRP RNAと同様に、ドメインIV構造の他にドメインI,II構造が存在することが明らかとなった。ドメインIV領域にはSRP54の相同因子であるFfhや蛋白質伸張因子EF-Gがそれぞれ独立に結合することを見いだしているが、本研究において、枯草菌SRP RNAドメインI領域に対して高い結合特異性を持つ蛋白質としてヒストン様蛋白質HBsuを同定した。HBsuに対する抗体を用いた免疫沈降実験により、HBsuとEfhはSRP RNAを介して細胞内で複合体を形成していた。また、HBsuの蛋白質分泌系における機能を調べるため、HBsuをIPTGで発現誘導可能な条件欠損異株を作成した。この変異株はIPTG非存在下で培養した結果、培養開始後5〜7時間で顕著な生育阻害を示した。変異株に枯草菌アルカリプロテアーゼ(AprE)、ペニシリン結合蛋白質5^(PBP5^),サイクロデキストリン合成酵素(CGTase)の各種シグナル配列と大腸菌β-lactamase(Bla)との融合蛋白質の発現プラスミドを導入、発現させ、Western blottingにより前駆体の蓄積の解析を行った。HBsu非存在下において培養開始後3時間ですでに、AprE-Bla、PBP5^*-Blaの約50%が前駆体として蓄積しており、CGTase-Blaでは約10%の前駆体の蓄積が観察された。これらの結果よりHBsuはFfh、scRNAと共にSRP様粒子を構成し、一群の蛋白質の分泌に関与していることが示された。
