食糧種子の難溶性蛋白質を特異的に分解する酵素とそのインヒビターの分子解析と応用

研究概要	コムギ種子CPであるグリアダインの活性組み換え体を得た。組み換えグリアダインはin vitroでの実験ではあるが、実際にコムギ種子の最も主要な貯蔵タンパク質であるグリアジンを分解した。グリアダインは発芽過程において貯蔵タンパク質(グリアジン)のプロテオリシスに関与していることが明らかになった。コムギ食品特有のタンパク質(グルテン)の品種改良、あるいは食品加工過程、例えば製パン工程等での利用価値が大いにあると思われる。一方、加工工程中、グリアダインの活性を制御することが必要であり、そのために、安全でかつ特異的なインヒビターが必要であることからコムギシスタチンの検索を行った。小麦シスタチンはオリザシスタチンをプローブに用いてスクリーニングを行った。その結果、3つの小麦シスタチンcDNAクローンを獲得した。これらはいずれもフィトシスタチンファミリーに属するものであった。大豆種子中に見出されたソヤシスタチンはフィトシスタチンファミリーに属するものの、N末端が長いという特長があったが、小麦シスタチンには既知のフィトシスタチンから突出するような特徴的な構造は存在しなかった。これらのことから、獲得したcDNAがコードするシスタチンタンパク質はパパインファミリーに属するトリティカインおよびグリアダインを阻害し得るものと推察される。 GST-progliadainは弱酸性条件下で活性を有した。この活性は市販の卵白シスタチン、オリザシスタチンで強く阻害された。また、2種の組み換え体小麦シスタチンによっても同程度に強く活性が阻害された。これらの結果は、小麦由来のCPとシスタチンを用いて、タンパク質分解反応系とその制御系を工業的に応用できる可能性を提示するものである。
