プラスミノーゲン・アクチベーター・インヒビター活性阻害による線溶活性化機構の解析

研究概要	血液凝固系と、これによりつくられた血栓を溶解する線維素溶解(線溶)現象は密接に関連しており、凝固がおこると線溶系は強く活性化されることが知られている。最近我々は、本機構の一つとして、線溶活性の重要な調節因子であるplasminogen activator inhibitor type-1(PAI-1)を活性化凝固因子が不活性化することに上り線溶活性を促進するという新しい考え方を示しており、この機構の生理的な意味を明かにしようとするものである。本年度の研究において、凝固カスケードの最終産生酵素であるトロンビンはPAI-1活性を中和するが、内皮細胞上に存在しトロンビン活性を修飾するトロンボモジュリンの存在下では中和しないこと示し、トロンビンによるPAI-1活性中和を介した線溶活性の促進はトロンボモジュリンの存在下では起こらないことを可溶性トロンボモジュリンを用いて精製糸で示した。凝固に伴う線溶活性の促進は、凝血塊上で過剰に産生されたトロンビンにより惹起されるが、正常な血管内皮細胞上に発現しているトロンボモジュリンに結合したトロンビンはこの活性を持たず、線溶活性の促進に関与しないことが示された訳である。これは生理的な凝固と線溶の調節に重要であると共に、血管内皮細胞が傷害された時に血管内の過剰な凝固系の活性化に伴い線溶活性が増強するという播種性血管内凝固症候群(DIC)の発生機序にも関与しているものと考えられた。来年度以降の研究において、上記反応を血漿あるいは全血を用いたcrudeな系を用いて生理的な意味合いを検証したい。
