プラスミノーゲン・アクチベーター・インヒビター活性阻害による線溶活性化機構の解析

研究概要

血液凝固系と、これによりつくられた血栓を溶解する線維素溶解(線溶)現象は密接に関連しておりその機構の解析が本研究の目的である。最近我々は、本機構の一つとして、線溶活性の重要な調節因子であるplasminogen activator inhibitor type-1 (PAI-1)を活性化凝固因子が不活性化することにより線溶活性を促進するという新しい考え方を示した。
昨年度の研究において、凝固カスケードの最終産生酵素であるトロンビンはPAIー1活性を中和するが、内皮細胞上に存在しトロンビン活性を修飾するトロンボモジュリンの存在下では中和しないことを示し、トロンビンによるPAIー1活性中和を介した線溶活性の促進はトロンボモジュリンの存在下では起こらないことを可溶性トロンボモジュリンを用いて精製系で示した。また今年度の研究で、euglobulin clot lysis time(ECLT)がca++と燐脂質の添加により凝固系を活性化することにより短縮することがわかり、これはprothrombin欠乏血漿では認められないことから、生成トロンビンによるPAIー1の不活性化によるものと考えられた。更にこの短縮は可溶性トロンボモジュリンで延長することから、凝固に伴い線溶活性が促進することと、それがトロンボモジュリンにより解消されることがより生理的な条件下でも示された訳である。これより当該機構は生理的な凝固と線溶の調節に重要であると共に、血管内皮細胞が損害された時に血管内の過剰な凝固系の活性化に伴い線容活性が増強するという播種性血管内凝固症候群(DIC)の発生機序にも関与しているものと考えられた。
