プラスミノーゲン・アクチベーター・インヒビター活性阻害による線溶活性化機構の解析

研究概要

血液凝固系と、これによりつくられた血栓を溶解する線維素溶解(線溶)現象は密接に関連しておりその機構の解析が本研究の目的である。最近我々は、本機構の一つとして、線溶活性の重要な調節因子であるplasminogen activator inhibitor type-1(PAI-1)を活性化凝固因子が不活性化することにより線溶活性を促進するという新しい考え方を示した。
昨年度の研究において、凝固カスケードの最終産生酵素であるトロンビンはPAI-1活性を中和するが、内皮細胞上に存在しトロンビン活性を修飾するトロンボモジュリンの存在下では中和しないことを示し、トロンビンによるPAI-1活性中和を介した線溶活性の促進はトロンボモジュリンの存在下では起こらないことを可溶性トロンボモジュリンを用いて精製系て示した。また、euglobulin clot lysis time(ECLT)がCa^<++>と燐脂質の添加により短縮する反応も、生成トロンビンによるPAI-1の不活性化によることを示し、更にこの短縮も可溶性トロンボモジュリンで解消されることを示した。またCa^<++>によるECLTの短縮は活性化proteinCにより凝固系の活性化を阻害することにより消失することも示し、当該機構は生理的な凝固と線溶の調節に重要であることを示した。これらは血管内皮細胞が傷害された時に血管内の過剰な凝固系の活性化に伴い線溶活性が増強するという播種性血管内凝固症候群(DIC)の発生機序にも関与しているものと考えられた。
