受容体細胞内動態に基づく受容体理論の再検討

研究概要	発光クラゲより同定されたGreen FIuorescent Protein(GFP)は極めて強い蛍光を有し、組み換え遺伝子手法により比較的容易に機能を損なわず蛋白標識を可能とすることから、各種生物学研究において汎用されつつある。α1アドレナリン受容体をモデル系として、遺伝子改変技術、各種薬理ツールを用い、受容体蛋白の細胞内動態に関する定裏解析を目的とした。方法としては(1)受容体蛋白発現:組み替え遺伝子発現コンストラクトを用い、N端にエピトープを挿入し、C端にGFPを持つようなα1受容体を一過性に発現、更には安定に発現する細胞を樹立した。遺伝子導入細胞はCHO細胞並びにαT3細胞を用いた。いずれの細胞においてもN端エピトープ並びにC端GFPの修飾が、受容体機能(受容体親和性、シグナル伝達能)に影響がないことを、受容体結合実験、カルシウム測定などにより一過性発現細胞で確認をした(2)受容体蛋白細胞内局在の定裏:エピトープに対する抗体を用いたflow cytometryによる解析より、細胞表蔓に存在する受容体蛋白を認識、定裏できる実験系を確立した。また、C端GFPは、その蛍光をflow cytometry並びに蛍光顕微鏡により同定でき、受容体蛋白の発現裏、分布をリアルタイムでモニターできる。また、GFPとは異なる波長域の蛍光指示薬でエピトープに対する抗体を標識することにより、二重染色も可能である事を証明した。(3)受容体蛋白細胞内局在論の観察:上述の遺伝子発現コンストラクトを遺伝子導入し、いろいろな程度に受容体蛋白を発現する細胞を作成し、受容体機能が遺伝子操作により影響を受けていないことを確認した後、これら細胞の中で、現在まで知られている各種組織に於けるα1受容体発現裏と比較して、生理的範囲内にある細胞と、非常に大裏に発現する細胞の2群を選択した。これらの細胞を用いて、受容体蛋白の生成、移送、分解などの細胞内局在機構の薬理学的解析を行った。結論として、α1Bアドレナリン受容体の作動薬による細胞内方運動はCキナーゼに依存しており、アクチン重合が関与しているが、一方ソーティングにはそれらの機構は関与せず、これら2つの細胞内分布運動は全く独立した分子機構によることが明らかとなった。
