平滑筋細胞形質転換(分化・脱分化)に関わるシグナル伝達系の解析

研究概要	これまでのニワトリ砂嚢平滑筋細胞を用いた解析によりIGF-I/IGF-I受容体の下流に位置するphosphatidylinositol 3-kinase(PI3-K)及びprotein kinase B(PKB/Akt)を介したシグナル伝達系が培養平滑筋細胞の分化型形質維持に重要な役割を果たすこととを明らかにしてきた。昨年度の研究で脱分化を誘導する増殖因子により協調的に活性化されるERK及びp38MAPKの両mitogen activated protein kinase(MAPK)を介したシグナル伝達系が脱分化誘導に深く関わることを見い出し、平滑筋細胞の形質はPI3-K及びPKB/Akt系とERK及びp38MAPK系の両シグナル伝達系のバランスにより決定されていることを明らかにした。本年度はこれら砂嚢平滑筋細胞を用いて得た知見を血管平滑筋細胞に応用した。この結果、血管平滑筋細胞も砂嚢平滑筋細胞と同様にIGF-I刺激下では分化型形質が長期間維持されることを見い出した。このIGF-Iによる分化型形質維持はPI3-K阻害剤により著明に阻害された。また、この培養血管平滑筋細胞もPDGF刺激で速やかに脱分化するが、ERK、p38MAPKを介するシグナル伝達系の阻害剤であるPD98059,SB203580により脱分化誘導が完全に抑制された。以上の結果から、血管平滑筋細胞においても内臓平滑筋細胞で明らかにした同様のシグナル伝達系が平滑筋細胞の形質を決定していることが明らかになり、これまでなされなかった分化型血管平滑筋細胞の初代培養系を確立させた。さらに、mRNAサブトラクション法により分化型平滑筋細胞に特異的に発現する因子の検索を行い、ホモフィリックな細胞接着に関わるカドヘリン6Bが分化型平滑筋細胞に特異的に発現している因子のひとつとして同定した。カドヘリン6Bは血管及び内臓平滑筋細胞由来の平滑筋細胞を血清刺激で脱分化させた細胞では全く発現が認められないことから、平滑筋細胞の形質依存性に発現制御を受ける新たな分子マーカーになる可能性が示唆された。その機構として分化型平滑筋細胞でのcell-cell相互作用に関わると考えられる。
