酸化ストレスによるアポトーシス機構と酸化ストレス反応配列(ORE)結合因子の解析

研究概要	(目的) 酸化ストレスによる炎症、虚血再かん流障害、環境化学物質による癌、アレルギーなどの機構を解折るため、アポトーシスと生体防御蛋白の発現誘導の二側面から酸化ストレスシグナルの分子機構を明らかにすことを目的に研究を行った。 (成果) 1、アポトーシスシグナル分子機構の解析:チオール基酸化剤diamideにより、Jurkat細胞でミトコンリアからのcytochrome cの細胞質内放出とcaspase-3活性化が生じ、アポトーシスが誘導された。グルタチオン成酵素阻害剤BSOの前処理によりそのアポトーシスの誘導は促進された。従って、diamideによるアポトーシス導にはミトコンドリアからのcytochrome cの放出と細胞内還元環境の調節が重要であった(上田他)。また、B2とTRXの発現がHIV感染による酸化ストレスとアポトーシスに関連した(Israel博士との共同研究)。 2、生体防御シグナル分子機構の解析: (1)抗酸化物質TRXは酸化ストレスにより転写レベルで誘導されるTRXプロモーター上に鉄ヘム化合物ヘミンに対する反応を伝える20塩基の配列(hemin responsive element,HRE)同定した(1998年生化学会にて発表)。その配列はNF-E2/AP-1様配列を含み、さらに、MF-E2/AP-1以外の因子関与が考えられ、HRE結合因子のスクリーニングを行っている。(2) ダイオキシン類によるTRXの転写活性化XRE様配列、SP-1結合配列を含む30塩基が必要な知見を得た(未発表)。(3)TRXのプロモーター上の酸化スレス反応配列(ORE)の結合蛋白について解析した。ORE結合蛋白の一つを精製し、microsequencingを行ったとこ80kDaの蛋白のN末アミノ酸配列と一致した。その結合の特異性および意義について検討中である(1998年癌学にて発表)。また、OREに結合する別の因子についてyeast one hybrid法にてscreeningを行っている。
