DMAHP/Six5遺伝子の機能と筋緊張性ジストロフィーへの関与

研究概要	筋緊張性ジストロフィーの原因となるCTGリピート下流に存在するホメオボックス遺伝子DMAHP/Six5の病因へのかかわりを明確にするために、病変が観察される組織である骨格筋、心筋およびマウス胚において、マウスDMAHP/Six5転写開始点を決定した。初期胚(E11)特異的な最も下流に存在する開始点1箇所と各組織に共通な2箇所の開始点を同定した。これらの転写開始点はいずれも筋緊張性ジストロフィーの原因となるCTGリピートの位置よりも下流に存在するので、CUGリピート配列がDMAHPmRNAに含まれた結果スプライシングの異常を起こしたり、ポリグルタミン等を含む異常蛋白の翻訳が病因に関与する可能性が排除された。プロモーター領域の転写制御エレメントを、本遺伝子が効率良く転写されているP19EC細胞への一過性形質転換法にて解析した。3箇所の正の制御エレメントは、いずれもSp1/Sp3結合エレメントであり、2箇所の負の制御エレメントのうち、1箇所には新たな特異的な因子の結合が見い出された。これら転写開始点と転写制御エレメントはヒトとマウスとの間でよく保存されているので、ヒトの筋緊張性ジストロフィーの病態解明にマウス遺伝子を用いることの有効性が明かとなった。現在、DMAHP/Six5の標的遺伝子を明らかにするために、VP16や既知の転写抑制ドメインを有する蛋白質との融合Six5遺伝子を作成し、その過剰発現によって発現変動が観察される遺伝子の同定を進めている。
