脂肪酸代謝蛋白の発現からみたヒト癌細胞の生物学的特性

研究概要	1.ヒト正常細胞では脂肪酸合成酵素(FAS)は主にホルモン感受性細胞(乳腺、前立腺など)・脂質代謝の活発な細胞(皮脂腺、脂肪組織、II型肺上皮など)および胎児で増殖の盛んな細胞に発現している。 2.各種ヒト癌における(FAS)の発現肺癌(n=200)では127/200(64%)、胃癌(n=434)では270/434例(62%),乳癌(n=243)では全例(但し高発現は145例),食道癌(n=88)では85/88例(97%)、軟部肉腫(n=64)では20/64例(31%)にFASの発現が認められた。FASの高発現が生存率と有意に相関したのは乳癌と軟部肉腫のみであった。 3.乳癌株MCF-7のFAS-mRNAはエストロゲンあるいはプロゲステロン処理により速やかに増加した。エストロゲンの効果はタモキシフェンにより阻害された。 4.胃癌株8種のFAS-mRNAの発現と脂肪酸代謝に関わる蛋白・酵素(アシルCoA結合蛋白;アシルCoA脱水素酵素→β酸化系;リソリン酸アシル転移酵素→リン脂質合成系;N-ミリストイルアシル転移酵素-2→蛋白質アシル化)のmRNAの発現は非常に高い関連性を示した。 5.FASの特異的阻害剤セルレニンでFAS陽性胃癌株Takigawaを処理すると、胃癌株の細胞の増殖は抑制され、脂肪酸代謝関連蛋白・酵素の発現は大部分低下した。以上のようにFASは細胞の脂肪酸代謝・ホルモン感受性と関連が深く、また脂肪酸代謝系と密接に連動して発現していることが確認された。また、FASはおおむね細胞の増殖能(胎児および癌)と関連性をもって発現することが明らかにされた。
