間質組織の構造と機能に関する基礎的研究:神経細網細胞複合体に着目して

研究概要

本年度は、まず第一に、マウス骨髄以外の組織における、神経細網細胞複合体構造の存在の証明を、ヒト卵管膨大部粘膜固有層内の間質細胞間に分布する、神経終末や、ギャップ結合の分布を免疫組織化学や電顕法を用いて検討することにより行なった。第二に、間質組織内の基本的な構成要素である、fibroblastの性格づけを、さらに進めるために、ヒト乳腺組織の小葉内及び小葉間の間質線維芽細胞につき細胞表面における,collagen type IV、laminin、fibronectinの分布を免疫電顕法を用い明らかにした。
さらに、以下の研究協力者たちとの、共同研究も進行中である。
慶大医耳鼻科、増田正純との、共同研究では、ヒト内耳前庭部暗細胞領域の上皮下間質組織内に存する、メラノサイト間のギャップ結合に関してコネキシンタンパクの分布を共焦点レーザー顕微鏡や金コロイド免疫電顕法を用いて検討中である。
慶大医眼科、石田晋との、共同研究では、病的間質組織である特殊な肉芽組織様組織の系として、黄斑円孔網膜剥離に対する硝子体手術において摘出された網膜上膜内の細胞が媒介するコラーゲン線維の改変化の機転の解明を、超微形態学的検討を加え行なっている。
慶大医整形外科、鈴木禎寿との、共同研究では、胎児期の重要な始原的な骨一骨髄間質組織の系として、C3Hマウス胎児を用いた、大腿骨におけるTRAPを指標とした、osteoclast,osteoblastの分化について検討中である。妊娠マウスを開腹し羊水中にVitamin D3およびcalcitoninを注入し、その後胎児太腿骨軟骨原基を軟骨膜ごと摘出し、観察する方法を用いて、骨形成期におけるVitamin D3およびcalcitoninの影響につき、電顕像を中心にTRAP酵素組織学的手法を加え検討している。
