研究概要	本年度の研究により以下の結果が得られた。 (1) Csk過剰発現系については、様々な機能変化が生じるメカニズムを解明する目的で、srcファミリーのチロシンリン酸化経路以外のシグナル伝達経路を検索した結果、MAPK(p44/42)、p38の活性が亢進していることが判明した。この変化と昨年明らかにした、LPS刺激後のアポトーシス細胞数の増加やアラキドン酸代謝産物(PGE_2など)の増加などが関連する可能性が示唆された。 (2) アログラフト炎症因子(AIF)-1をクローニングし(AB013745)、融合蛋白質を作製した。これを免疫原として、単クローン抗体を樹立し、組織分布、動脈硬化巣での発現などを検索したところ、正常マウスにおいてはAIF-1は、脾臓の単核球、精巣の精細胞、動脈硬化巣にあっては内膜下に遊走し泡沫化しつつあるマクロファージに、発現することが判明した。 (3) AIF-1の機能を解析する目的で、過剰発現細胞をマウスマクロファージ細胞株RAW264.7で作製し、細胞形態・モノカイン産生が大きく変化することが判明した。 (4) MCP-1 Tgmで接触性過敏反応が増強しているのは、MCP-1の作用によって、皮膚からのランゲルハンス細胞の遊走が亢進しているためであることが判明した。 (5) MCP-1 Tgm でLPS投与によって辺縁帯マクロファージ亜群がアポトーシスに陥るのは、同Tgmにおいて炎症性サイトカインが高値であることに関連していると考えられた。 (6) 動脈硬化症マウスモデル(ApoE-/-)を用いて、骨髄細胞側に体現される、動脈硬化症発症抵抗性遺伝子が存在することが判明し、ある種のヒト高脂血症とそれに随伴する動脈硬化症において骨髄移植が有効な治療法足り得ることが判明した。
