HTLV産生細胞の各種抗原発現に関する免疫電子顕微鏡的研究

研究概要

本年度は、HTLV-I、-II産生細胞株に関連し、他のT細胞株をも含め、各種マーカーの発現の有無をまず光顕にて検討した。用いた細胞はヒトHTLV-I、-II産生細胞の他にTALL-1、-2、DL-40のT細胞株、DL-95、DL-110のCD30陽性細胞株、更にprimary efrusion lymphoma(PEL)由来のHHV-8産生細胞KS-1とそのTPA処理細胞および巨核球細胞株MKPLについて主要なマーカーを検討した。HTLV-Iは、IL-2Rβは陰性、CD44は陽性、CD11a(LFA-1a)は陰性という結果を得た。HTLV-IIa、IIbについても同様であった。TALL-1、-2とDL-40はCD4が陽性であり、TALL-1、-2はCD5も陽性であった。DL-95、DL-110はCD30の他に各々CD4、CD19も陽性であった。KS-1とTPA処理KS-1については、KS-1ではHHV-8粒子の出現は極端に少なく、核内のみに限られており、それらの微細構造の検討は困難であったが、TPA処理により多数の核内、胞体内外、粒子産生が誘発され、HHV-8の未熟粒子や成熟粒子の微細構造観察が可能であった(日本臨床電顕学会発表・大朏ら、1998)。このKS-1とTPA処理KS-1についてもマーカー検索を行い、共に、CD45、CD20、CD5、CD30、HLA-DR陽性という結果を得た。これら各種細胞の光顕的に陽性と考えられたマーカーについて目下電顕レベルでの細胞内局在について検討中である。これらマーカーは必ずしも細胞膜表面に発現するのみでなく、既検索結果では例えば、CD3は細胞膜表面と核周囲腔に、またTCRα/βは細胞膜表面に陽性となるが、TCRαとTCRβは核周囲を含む粗面小胞体に陽性であった。これらの発現状況からTCRα/β、そのα、β鎖についてはかなり知見が得られ発表してきた(Ohtsuki et al,Med Electron Microsc,1997)。従って様々な種類のT細胞株を主体とした細胞株における微細構造レベルでの抗原発現状況については興味深い所であり、平成11年度に詳細を検索予定である。一部は、日本電顕学会シンポジウム(大朏、1999)において発表予定である。
