HTLV産生細胞の各種抗原発現に関する免疫電子顕微鏡的研究

研究概要

HTLV-I、-II産生細胞株について、T細胞受容体(TCRα/β)、およびそのα鎖、β鎖について発現状況を検討し、特に新鮮ヒトリンパ球におけるそれらと比較検討した。まずHTLV-I、-II細胞では、CD25、HLA-DRは共に強陽性であり、CD4は、HTLV-I産生長期培養株では減弱していたが、CD3は一部で残存しており、細胞膜表面のみでなく、核膜周囲腔や粗面小胞体に陽性であった。TCRα/βは、HTLV-Iや-II産生細胞株では細胞表面には、ごく一部で認められたのみで殆ど陰性であった。TCRα鎖は、新鮮リンパ球では、核膜周囲腔や粗面小胞体にTCRβ鎖と共に陽性であったが、培養細胞では、TCRα鎖の減弱、消失やTCRβ鎖の遺残をみ、表面に陽性であったTCRα/βも陰性化した。HTLV-I、-II産生細胞では、TCRβ鎖が一部で陽性であったのみで、TCRα/β、TCRα鎖は共に陰性化していた。これらの結果は、HTLV-I、-II産生細胞では、T細胞受容体複合体(TCR・CD3)が不完全であり、成熟したT細胞としての機能を有さない機能不全状態にあることを示唆している。また、TCRα/βの細胞表面からの消失は、新鮮リンパ球でも明らかであったようにα鎖の消失に続く二次的なものであることが示唆された。またPrimary effusion lymphoma(PEL)由来のKS-1細胞とそのTPA処理細胞とで行ったHHV-8の超微形態所見では、TPA処理において核内のみでなく胞体外に放出される未熟ないし成熟粒子が多数観察され、外径250μmをこえる巨大粒子や発芽粒子も確認された。TPA未処理細胞では核内に少数の粒子をごく少数の細胞で認めたのみであった。(Ohtsuki et al,Med Electyron Microsc,1999)。これらのデータはHHV-8粒子の微細構造を明らかにした責重なデータである。
