γδT細胞の発達分化と生体内生理的機能の解明

研究概要	マウス腸管の上皮細胞層には膨大な数のγδT細胞が分布する(γδ-IEL)。γδT細胞の生体内生理的機能は未解明であるが、γδ-IELのほぼすべては特異なCD8αα分子を発現し、胸腺非依存性に腸管粘膜局所で発達分化することが明らかにされている。我々は通常マウスから精製したαβ-IELとγδ-IELがin vitroでキラーT細胞(CTL)活性を発揮するのに対して、無菌マウスではαβ-IELのCTL活性のみが消失し、γδ-IELのCTL機能は保たれることを明らかにした。近年、T細胞抗原レセプター(TCR)遺伝子β鎖ミュータント(β^<-1->)マウスとTCR-α鎖ミュータント(α^<-1->)マウス双方のIELは、ほぼすべてγδ-IELであるにもかかわらず、α^<-1->マウスが16週令頃より高頻度に炎症性腸疾患(IBD)を発症することが明らかにされている。我々は8週令から20週令までのβ^<-1->マウスとα^<-1->のγδ-IELについて、そのCTL活性と抗γδ-TCR単クローン抗体刺激によるin vitro IFN-γ産生能を検索した。その結果β^<-1->マウスγδ-IELのCTL及びIFN-γ産生機能は著しく低下し、正常wildーtypeマウスγδ-IELよりはるかに減弱するのに対してα^<-1->マウスγδ-IELの両機能はIBD発症以前より著明に亢進することが判明した。γδ-IELの発揮するCTL活性は腸管上皮細胞を傷害(標的抗原は未解明)して排除することによって上皮細胞の新生をうながすことが提示されている。今回得られた知見によって、α^<-1->マウスの腸管上皮細胞に、弱齢期より異変が引起こされていることを示唆するとともに、γδ-IELがIBD発症防御的に働く可能性が提示された。
