研究概要	過去数年にわたる研究からわれわれはマスト細胞が粘膜免疫において重要な役割を担っていることを明らかにしてきた。このことから、マスト細胞の機能を遮断することによりさまざまな免疫学的効果がえられることが期待された。そこで、マスト細胞へのIgEの感作を可等型IgEレセプター(Fc_εRI)により阻止することによるマスト細胞の機能抑制を考えた。可溶型IgEレセプター蛋白そのものの大量精製は膨大な費用と手間を要するので、発現プラスミドによる遺伝子治療の手法を用いた。古典的即時型アレルギーモデルであるラット受身皮膚アナフィラキシー(PCA)反応をマスト細胞の機能の指標として用い検討した。 1)transientの発現系より得た培養上清から可溶型Fc_εRIα鎖に相当する分子量の蛋白を免疫沈降できること、2)その培養上清はIgEをin vitroで吸収し、PCA反応を抑制したことからわれわれの作成した可溶型マウスFc_εRIα鎖遺伝子発現プラスミドはin vitroで可溶型Fc_εRα鎖蛋白を産生することが示された。また、IgE抗体皮肉感作24時間前にIgE感作部位直下皮下にプラスミドを注射したところ、投与プラスミドの用量依存的にラットPCA反応の抑制した。以上より、われわれの作成した可溶型Fc_εRIα鎖遺伝子発現プラスミドはin vivoおよびin vitroで生理活性を示す可溶型Fc_εRIα鎖を産生することが示された。発現プラスミド直接投与によりマスト細胞の抑制が可能であることを示すとともに、可溶型Fc_εRIα鎖遺伝子を導入することによるアレルギーの遺伝子治療の道をひらくものと考えられた。
