B型肝炎ウイルスキャリアのワクチン療法に果たす樹状細胞の役割

研究概要

B型肝炎ウイルス(HBV)の持続感染において樹状細胞が重要な役割を果たし、B型肝炎ウイルス遺伝子トランスジェニック(HBV-Tg)マウスでは樹状細胞の抗原提示能が障害されていることを証明してきた。このマウスにHBsワクチンを投与するとHBVの増殖が停止しHBs抗原とHBe抗原が陰性化した。さらにワクチン治療によりマウス樹状細胞機能が回復した。
HBV Tgマウスに対するHBVワクチンの作用機序を明らかにする目的で、樹状細胞の抗原提示能と表面マーカーの変化及びサイトカイン産生能について検討し、ワクチン療法が有効であったマウスではDC機能が改善し、その機序はDCのMHCclassII,CD86の発現亢進とIL-2,IL-12,TNF-αの産生亢進が認められた。
そこで治療前の樹状細胞機能によりワクチン治療の効果が予測できるか検討した。
さらにHBV-DNAワクチンについても検討した。
ワクチンが有効であったマウスはワクチン治療前に樹状細胞機能をKLHに対する反応で検討すると反応性の高いDCが存在し、より活性化された樹状細胞を持つマウスのみがワクチンに反応しワクチンの効果予測が可能であった。
DNAワクチンをHBVトランスジェニックマウスに投与した実験では1回の投与のみで通常のHBVワクチン以上の効果が得られた。DNAワクチンは通常のHBVワクチン投与時以上に樹状細胞機能の改善をもたらしHBV Tgマウスに効果を及ぼしている可能性が示唆される。
