インターフェロンの肝発癌抑制効果の解析と臨床応用への基礎的研究

研究概要

癌細胞増殖抑制効果を指標にしてインターフェロンの肝癌細胞に対する作用を検討した。抑制効果の高い(高感受性)細胞と、低感受性細胞を用いて、シグナル伝達物質であるinterferon regulatory factor(IRF)-1の遺伝子変化と発現量の違いをみた。高感受性細胞ではIRF-1mRNAの発現が高く、低感受性では発現量が少なかった。前年度の研究から、IRF-1genomeにrearrangementをきたす違いが考えられたため、BamHI siteの違いをintron部に追及したところ、intron8の部位に低感受性株にのみ認められるBamHI siteを見いだした。健常人ボランティア末梢血DNAの検討から、この変位は個体に見られるsingle nucleotide polymorphysm(SNP)であることが判明した。さらに、プロモーター部位を増幅し、クローニング後sequenceを行ったところ、データベース上の-500bpまでの位置に4点のSNPを見いだした。このSNPを含む約500bpのプロモーター部位をベクターに組み込み、高感受性株、低感受性株それぞれに導入して、ルシフェラーゼアッセイを行ったところ、プローモーター活性に有意な差が認められた。すなわち、インターフェロンに対する薬剤感受性を規定するSNPであることが考えられた。現在、健常人のリンパ球を用いてSNPとインターフェロンの感受性の関係について検討中である。
