アレルギー性気道炎症でのTリンパ球の局所集積・活性化における気道上皮細胞の役割

研究概要	気管支喘息をはじめとするアレルギー性気道炎症では、Tリンパ球とくにTh2タイプのリンパ球の役割が重視されている。しかしながら、このTリンパ球の気道局所への動員機構や活性化の機序についての知見はきわめて乏しいのが現状である。本年度はまず、ヒト気道上皮細胞から産生されるTリンパ球へのケモカインとして、RANTESとMIP-1を中心に検討し,前者はTh2タイプのリンパ球が産生するIL-3+GM-CSF刺激で誘導されそのmRNAもRT-PCR法にて発現の増強を確認した。また、最も強力な喘息治療薬である副腎皮質ステロイドはこの過程を用量依存的に抑制した。しかし、後者の発現は試みたいづれの刺激によっても認められなかった。さらにthymus and activation-regulated cytokine (TARC)のヒト気道上皮細胞での遺伝子発現を同じくRT-PCR法にて確認した。今後そのin vitroでの発現調節機構とin vivoでの発現状況を明らかにしたい。また、気道上皮細胞のTリンパ球活性化における意義については、主にヒトTリンパ球のヒト気道上皮細胞への接着の検討を進めており、IFNγおよびTNFαによる増強効果とその際の気道上皮側でのICAM-1(CD54)の発現増強、さらに抗ICAM-1中和抗体による接着過程の抑制傾向を認めた。また、その際Tリンパ球からのIL-10産生・遊離も抑制される傾向を認めた。このように、リンパ球の気道上皮への接着自体がリンパからのTh2タイプのサイトカイン産生を調節する可能性につき、今後さらに遺伝子発現も含め検討する予定である。
