脊髄運動ニューロンにおけるglycationと最終糖化産物-oxidative stressとのsynergism-

研究概要	AGEs生成を促進するMG、3-DGが培養大脳ニューロン、脊髄ニューロンに対して用量依存性の毒性を示し、大脳ニューロン、脊髄非運動ニューロンに比較して脊髄運動ニューロンのsusceptibilityが高かった。大脳ニューロンでは活性酸素種(ROS)の関与が示唆され、N-acetylsysteine(NAC)が保護作用を示した。NACによる保護率は運動ニューロンで高く、非運動ニューロンに比較してGSH systemが不十分な可能性が示唆された。GSH level自体は運動ニューロンと非運動ニューロン間で差は認められず、その代謝、利用率などによる脆弱性が推測された。AGの保護作用は共処置でのみ認めたことから、MG、3-DGへの直接作用と考えられた。ヒト剖検ALS脊髄でglycation初期生成物とAGEsの存在を確認した。CMLは主に運動ニューロン胞体とグリアに存在した。しかしCMLはlipidperoxidationを反映していると考えられる。培養細胞にてNO供与体でCMLは産生増加しnon-CMLに変化が認められなかったことはこれを支持する。Glycation初期生成物である1-HL、FLは主にspheroidsに染色性を認めており、その構成成分から標的としてneurofilamentの可能性が考えられ、持続的かつ常時新しいglycationがおこっていることが示唆された。それに引き続きglycation後期反応が進んだ結果としてnon-CML AGEの存在が確認された。Non-CML抗体はspheroidsに加え、運動ニューロン胞体にも染色性を認めており初期生成物の結果と解離していた。抗体の感度や標的蛋白の違いを反映していることが推測される。以上、ALSにおけるglycationの関与をin vitro、in vivo両面より確認した。
