グルタミン酸受容体自己抗体による中枢神経系障害の早期診断・治療

研究概要	われわれは、グルタミン酸受容体に対する自己抗体による中枢神経系障害を早期に診断し、早期治療に導くための研究を行ってきた。本年度は以下のような研究成果が得られている。 (1) グルタミン酸受容体に対する自己抗体検出システムの確立グルタミン酸受容体の内のε2を発現するテトラサイクリンシステムのレポーター遺伝子を、テトラサイクリンシステムのトランスアクチベーターをステイブルトランスフェクションした細胞(3T3-TA)にエレクトロポレーションし、3つのクローンがε2-cDNAを組み込んでいる事をPCRにて確認した(3T3-TA-ε2)。3T3-TA-ε2のホモジネートをε2抗体を用いたウエスタンブロットにて解析したところ、ε2蛋白の発現を確認できた。このことより、抗ε2抗体のスクリーニングが可能となった。 (2) グルタミン酸受容体に対する自己抗体の検出現在、3T3-TA-ε2を用いてepileptia partialis continuaを呈したてんかん患者さんの血清・髄液中の抗ε2抗体を、IgG,IgM,IgA分画に分けて検索中であるが、IgM分画に陽性所見かと思われるバンドのある患者さんがあり再現性を見ているところである。 (3) テトラサイクリンシステムのグルタミン酸受容体発現用レポーター遺伝子(ε2、δ2以外)の構築 ε1、ε3、ε4、ξ1のcDNAを現在調製中で、制限酵素サイトが確認出来次第、グルタミン酸受容体発現用レポーター遺伝子を構築する。次年度の計画現在δ2-cDNAを3T3-TAにトランスフェクションし、ピュウロマイシンにて選択中であるので、ステイブルクローンが得られ次第、抗δ2抗体を用いてδ2蛋白の発現を確認し、患者血清のスクリーニングに入りたい。また抗ε2抗体のスクリーニングを幅広い患者で行い、他のグルタミン酸受容体に対する自己抗体検出システムの確立を急ぐ予定である。
