食物抗原によるアトピー性皮膚炎発症のメカニズム及び治療法に関する基礎的検討

研究概要	平成11年度はヒトナイーブT細胞のサイトカイン産生能の獲得機構を明らかにする目的で以下の実験を行った。 1)昨年度ヒト臍帯血CD4+T細胞を用いた実験により、IL-7によって誘導されるIL-4産生細胞はすべてCD31陰性であることを明らかにした。従って、CD31の発現調節を明らかにする目的で、CD31陰性細胞を臍帯血より分離し、IL-4とIL-7で培養した。IL-7は最初に存在したCD31陰性細胞を維持しこの分画にIL-4産生能を誘導した。IL-4はCD31を細胞表面に誘導する効果があり、CD31陽性細胞にはIL-4産生能は誘導されなかった。CD31を介する刺激がIL-4産生能に抑制的に関与している可能性が示唆された。 2)IL-4やIL-7により抗原刺激の非存在下でIFN-γ産生能を獲得したナイーブT細胞を用いてIFN-γ産生の分子機構を検討した。抗原刺激の非存在下でIFN-γ産生能を獲得したT細胞のIFN-γ遺伝子上流域はメチル化されており、DNase I hypersensitivity assayでも通常のメモリーT細胞のIFN-γ産生に使用されているプロモーター領域が使用されていない可能性が示唆された。従って、ヘルパーT細胞のIFN-γ産生機構はその分化様式に応じて多様性が認められた。 3)食事アレルギー患者の末梢血より単核球を分離し、該当抗原で刺激培養上清中のサイトカインを測定することにより、末梢血中の食物抗原特異的T細胞の頻度を測定する実験系を検討した。この実験系では測定するサイトカインが微量のため、加える抗原の中に微量のエンドトキシンが混入していても測定系に影響を与えることが明らかになり、現在再検討中である。
