酵母機能アッセイ法を用いたヒト皮膚発癌におけるP53癌抑制遺伝子の役割の解明

研究概要	本研究の目的は、簡便にかつ鋭敏にp53遺伝子変異を検出できる酵母p53機能アッセイ法を用い、種々の皮膚腫瘍におけるp53遺伝子の異常を検出することで、皮膚発癌におけるp53遺伝子異常の意義を明らかにする事にあった。平成11年度に、50例の皮膚腫瘍(黒色腫10例、基底細胞癌12例、有棘細胞癌5例、ぺージェット病1例、エックリン癌1例、毛包癌3例、日光角化症1例、ボーエン病5例、皮膚線維腫1例、母斑細胞性母斑11例)について本法によるp53遺伝子変異の検出を行ない、その結果、10例中2例の黒色腫、12例中3例の基底細胞癌、5例中3例の有棘細胞癌、1例中1例のエックリン汗器官癌、3例中3例の毛包癌、5例中2例ボーエン病をp53遺伝子異常陽性と判定した。1例のページェット病、1例の日光角化症、1例の皮膚線維種、11例の母斑細胞性母斑では全て陰性であった。DNA sequence法については、実験手技の確立に手間をとったため、現時点で数サンプルについてのみ施行できただけであるが,酵母p53機能アッセイ法陽性のサンプルでp53遺伝子変異が確認された。また、酵母機能アッセイを施行した50例のうち、35例のパラフィン切片を用いて、p53蛋白の発現を免疫組識化学的に検討した。p53蛋白の過剰発現がみられた8例中6例が酵母機能アッセイで陽性を示したが、2例は酵母機能アッセイ陰性であった。逆にp53蛋白陰性であった27例中26例が酵母機能アッセイ陰性で、残りの1例は酵母機能アッセイが陽性であった。以上、2年にわたる研究の結果、本実験を手技的に確立でき、p53遺伝子変異を検出する上で、本法が比較的簡便で感度の高い手法であることを確認し得た。今後さらに症例を蓄積し、酵母機能アッセイ法、免疫組識化学、DNAシーケンスの3者を用い、各種皮膚腫瘍の発癌におけるp53遺伝子変異の全容を明らかにしたい。
