トロンボポエチン/c-mpl系を介した増殖・分化・腫瘍化のシグナル伝達機構の解析

研究概要	トロンボポエチン(TPO)は巨核球系細胞の増殖と分化において最も重要な造血因子である。TPOは、巨核球系細胞表面に発現するTPO受容体c-mplに結合することによりSTATs、ras、PI-3Kなどの細胞内シグナル伝達分子を活性化することが報告されているが、TPOよる増殖・分化誘導にどのシグナルが重要であるかは明らかではない。本研究では、まず、ヒトのIL-3依存性赤白血病株F-36Pにc-mplを導入した。このうちの一つのクローンF-36P-mplにおいて、TPOは一過性に増殖反応を誘導し、その後ほとんどの細胞に成熟巨核球への分化を誘導した。F-36P-mplにdominant negative(dn)-STAT1,3,5,dn-rasをLac一inducible Systemを用いてinducibleに発現させ、TPOによる増殖、分化の誘導がこれらの分子によって阻害されるかどうか検討を行った。その結果、TPOによる増殖誘導はdn-STAT5とdn-rasによって各々約30%抑制された。また、分化誘導はdn-rasによってのみ阻害された。更に、活性型rasをF-36P-mplにinducibleに発現させるとTPO非存在下でも成熟巨核球への分化が誘導された。活性型rasの発現時間と巨核球への分化誘導効果との関連を検討すると活性型rasを約24時間発現させることが巨核球への分化誘導には必要かつ十分であると考えられた。rasのシグナルはTPO以外にもIL-3やSCF、EPOなどの各種の造血因子によって活性化されるが、TPOは、IL-3と比較すると持続性に約24時間にわたってrasを活性化した。これらの結果から、TPO特異的に誘導される持続性の長いrasの活性化が巨核球への分化誘導には重要であると考えられた。
