好酸球分化に関与する新規転写因子のクローニングとその機能解析

研究概要

好酸球は、アレルギー疾患・寄生虫疾患・悪性腫瘍など広範囲な病態において重要な役割を果たしている骨髄由来の顆粒球系細胞である。この好酸球の骨髄からの分化を決定する遺伝子を明かにするため、好酸球に特異的に発現しているEosinophil major basic protein(MBP)の遺伝子発現の調節機序の研究を計画し以下の成果を得た。
(1) MBP遺伝子は32kb離れてエクソン1とエクソン9の5'上流に各々P1及びP2の二つのプロモーター領域が存在するが、好酸球に特異的なプロモーターはP2である。一方、P1プロモーターは、trophoblastに特異的なプロモーターであることが示唆された。
(2) P2プロモーターの解析から、-117〜-67bpに促進的に作用するシスエレメントが存在し、その中でGATA結合領域(-76〜-71bp)がMBP遺伝子発現に重要な領域であることが明かになった。
(3) GATA結合領域(-76〜-71bp)には、GATA-1及びGATA-2が結合することかゲルシフトアッセイで証明された。さらに、この領域に対する結合親和性はGATA-1の方がGATA-2に比して高かった。機能的にはGATA-1かMBPプロモーター活性を促進的に増強させ、一方、GATA-2はMBPプロモーター活性を競合的に抑制した。
(4) GATA結合領域(-76〜-71bp)の7bp上流にC/EBP結合領域が認められ、そのうちC/EBPβがGATA-1と強調してMBP遺伝子発現を増強させることが明かになった。
以上の結果、好酸球分化においでGATA-1及びC/EBPβが重要な転写因子であることが示唆された。
