腎糸球体上皮細胞の培養系の確立

研究概要	多くの研究室で報告されている糸球体上皮細胞(VEC)の培養細胞の性質は、in vivoのVECの形質と大きく異なり、敷石様配列をとったものがほとんどである。この細胞の由来については、脱分化したVECであるとする意見とPEC由来の細胞であるとする意見が対立している。我々は、このような混乱を招いた原因のひとつとして、ボウマン嚢の被っていない単離された糸球体(G)から、ほとんど細胞が培養されてこないことに注目した。従来の糸球体単離のためのsieving methodには、腎組織片をメッシュに押しつける操作があり、これにより糸球体表面のVECが傷害を受け、死滅することが予想される。そこで、単に腎皮質を鋭いナイフで細切するだけで、単離されてくる糸球体を位相差顕微鏡下で拾い集めて、培養を試みた。その結果、Gからの細胞が生えだす率は著明に改善した。その細胞形態は、長い細胞突起を伸ばした不規則なものであり、敷石様配列の細胞とは明らかに異なっていた。これらは、Thy-1、vWF、RECA-1が陰性であり、メサンギウム細胞、内皮細胞とは異なるため、VEC由来であることが確かめられた。さらに、我々は、pan cadherinに対する抗体染色がPECに陽性でVECに陰性であることを発見した。これで培養細胞を検討すると、G由来の細胞は陰性であり、ボウマン嚢を被った糸球体から生えだす敷石様の細胞は陽性であった。以上のことから、敷石様の細胞はVEC由来ではなく、PEC由来、または尿細管が混入している場合は、尿細管上皮由来であることが明らかとなった。これらの系を使って、in vivoでのVEC特異マーカーを検討したところ、マーカーのいくつかのものは、敷石様細胞にも陽性になることが示され、in vivoで特異的であるものが培養系でも特異的であるとは限らないことが分かった。
