レプチンの胎生期における機能の発生工学的解析

研究概要	レプチンの胎生期における機能を明らかにするため、前年度までに胎生期におけるレプチンの作用部位および時期を特定する目的で、レプチンレセプター(Ob-Rb)mRNAの発現パターンを胎仔および新生仔マウスにおいて検討した。その結果、中枢神経系において、RT-PCR法では既に胎生10.5日に、in situハイブリダイゼーション法では胎生11.5日より発現が認められた。よって、マウスの胎仔発生にレプチンが関与していることが考えられた。そこで、今年度はまずレプチン欠損(ob/ob)マウスと野生型(B6J)マウスの胎仔の形態を比較し、全身および中枢神経系の発生におけるレプチン欠損の影響を検討した。胎生16日および18日では、ob/obおよびB6Jマウス胎仔の頭殿長および体重に差は認められなかった。しかしながら、中枢神経系のニッスル染色切片を観察したところ、胎生16日のob/obマウスでは、B6Jマウスと比較して間脳および中脳が小さかった。胎生18日の胎仔については、現在観察中である。次に子宮外発生法を用いて、ob/obマウス胎仔脳室内にレプチンを注入し、全身および中枢神経系の発生に関するレプチンの機能を検討した。なお、対照群には溶媒(クエン酸ナトリウム+リン酸緩衝液)のみを注入した。胎生14日にレプチンを1μgあるいは200ng投与し、胎生16日に観察したところ、対照群と比較して前者では頭殿長および体重の減少が認められ、後者では体重のみの減少が認められた。一方、胎生16日にレプチンを投与し、18日に観察した群では、1μgおよび200ng投与群とも対照群との間で頭殿長および体重に差は認められなかった。レプチン投与による中枢神経系の発生における形態学的変化については、現在解析中である。以上より、胎仔の発生、特に頭殿長、体重および中枢神経系の発生にレプチンが関与していることが示唆された。
