動脈硬化進展におけるVEGFの役割に関する研究

研究概要	本助成により、vascular endothelial growth factor(VEGF)は酸化的変性低比重リポ蛋白(Ox-LDL)による内皮細胞障害を2種類の機構(一酸化窒素依存性、細胞内グルタチオン依存性)によって抑制することを明らかにした。昨年度はVEGFのOx-LDLによる内皮細胞障害抑制効果が、VEGFの2種類の受容体、Flt-1ならびにKDR/Flk-1のいずれの受容体を介しているかを明らかにするために、それぞれの受容体の強制発現系を作成し検討した。しかしながら、この細胞のいずれもVEGFのOx-LDL毒性抑制効果を認めず、この実験系に疑問が残った。今回、Flt-1,KDR/Flk-1の特異的中和抗体を使用し、VEGFの内皮細胞防御効果に関与する受容体の同定を試みた。Flt-1の特異的中和抗体はVEGFのOx-LDLによる内皮細胞障害抑制効果に影響を与えなかったが、KDR/Flk-1抗体は濃度依存性にVEGFの障害抑制効果をブロックした。さらにKDR/Flk-1抗体はVEGFによる内皮細胞内のグルタチオン発現の増加を濃度依存性に抑制した。以上より、少なくとも細胞内クルタチオンを介する機構では、VEGFは内皮細胞のKDR/Flk-1受容体を介して、Ox-LDLによる内皮細胞障害を抑制していることが明らかとなった。この結果は以前Flt-1のリガンドであるplacenta growth factorにはOx-LDLに対する内皮細胞障害抑制効果が無いこと、さらに細胞内グルタチオン発現にも影響が無い結果と一致する。
