研究概要	膵B細胞のグルコース応答性インスリン分泌には、時相の早い第I相とそれに続く第II相が存在する。膵B細胞で発現する解糖系の初段階の律速酵素、グルコキナーゼ(GK)には、ヘキソース特異性、アノマー特異性、基質Kmなどにセンサーの特質が認められる。次の段階を律速するホスホフルクトキナーゼ(PFK)はGKと異なり、高度なアロステリック調節機構を有し、細胞内水素イオン濃度、クエン酸・ATP濃度など解糖の結果蓄積する反応産物による阻害をアイソザイム別に受ける。よってPFKステップがそのアイソザイムごとの特色ある反応を触媒することがグルコース応答性インスリン分泌の第I相・第II相形成に寄与することが考えられる。本年度は、細胞生物学的手法によって本研究の目的とする仮説を部分的に検証する準備を行った。 1. ネオマイシン選択性を有するベクターであるpCXN2にマウスPFK-L、Mの開始コドン周辺約300塩基程度のcDNA断片を逆方向に挿入したアンチセンス発現ベクターを構築した。 2. これを用いてMIN6細胞からstable transformantを作成中である。 3. 次年度には樹立した細胞株を用いてグルコース応答性インスリン分泌の時相変化、解糖代謝の変動、細胞内カルシウム濃度の経時的変化などを分析する。これらの結果を総合評価し、PFK-Mノックアウトマウス作成の理論的基盤を得た後、ターゲッテイングベクターを構築していく。
