インスリン標的細胞におけるGFATを介した糖毒性発現の分子機構とその治療

研究概要

(これまでの経過)
rat glutamine:fructose-6-phosphatase(GFAT)cDNAを発現ベクターpCXN2に挿入したpCXN2-GFATを作製した。このvectorをlipofection法(transfectam)でMIN6にtransfectionし、geneticin(G418)で選択し、約100個のstable cloneを得た。それらのcloneで実際にrat GFATcDNAが発現しているか確認するため、RT-PCR法およびNothern blot法を行ったところ、少なくとも5cloneで発現を確認した。
(今後の方針)
1) これらのcloneを用いて、GFAT活性、グルコース刺激インスリン分泌を測定し、その相関について調べる。
2) GFATの阻害剤としてazaserineが知られている。GFAT過剰発現によるグルコース応答性インスリン分泌抑制がazaserine添加により解除できるか検討する。
3) GFATはglutamineおよびfructose-6-phosphateよりglucosamine-6-phosphateを形成する。また、glucosamineはhexokinaseによりglucosamine-6-phosphateとなることが知られている。そこで、様々な濃度のglucosamine添加によりグルコース応答性インスリン分泌が抑制されるか検討し、GFAT過剰発現細胞で得られた結果と比較する。
