脂質輸送におけるMTPの役割とMTP蛋白の発現制御機構の解明

研究概要	我々はスンクスにおける脂肪肝発症メカニズムの検討から、ACAT(Acyl-CoA Cholesterol acyl transferase)活性がMTP(microsomal triglycerides transfer protein)活性の制御因子になっている可能性を考慮して以下の実験を行った。まず、マウス肝臓より精製したmRNAを用いて、RT-PCR法にてACAT-1のcDNAの全長をクローニングした。これをシャトルベクターpAdCMV Link.1のMCS(multiple cloning site)に挿入した(pAdCMV mACAT)。293細胞にpAdCMVmACATをtransfectionし、このプラスミドがACAT蛋白を発現すること、そして発現されたACAT蛋白がACAT活性を有することを確認した。続いて、作製したvectorをリニアライズした後AdCMV-LacZ DNAとともに293細胞にco-transfectionしてACATgeneを含むアデノウイルス発現ベクター(AdCMV-mACAT)を作製した。またシャトルベクターと同様にその効果(発現)については293細胞へのInfectionにて確認した。次にスンクス肝臓へのAdenovirusを用いた導入効率がどの程度であるか調べるためにAdCMVlacZを用いた検討を行い、以下の結果を得た。1匹あたり3.5×10^<11>particlesの投与量での肝細胞への遺伝子導入率は約15%程度であった。またそれ以上の量を投与した場合ではスンクスが耐えられず、静注後約2日目に、遺伝子が十分発現する前に死亡した。そこで、in vitroで培養肝細胞への遺伝子導入を試みたところほぼ100%の細胞に遺伝子を導入できた。現在、導入細胞におけるMTP活性、VLDLの分泌能について検討を加えている。
