HLA-A24陽性食道癌ペプチドワクチンの開発:ペプチドによる末梢リンパ球からのCTLの誘導

研究概要	これまでに樹立した食道癌患者由来上皮性癌特異的キラーT細胞株(KE4-CTL)により認識される5つの抗原をSART-1〜SART-5をクローニングし、それらの各種癌における発現、新規の場合は生物学的機能及び、CTLにより認識されるエピトープ(ペプチド)、さらにはCTL誘導能可能なペプチドの決定等々を実施した。以下その概略を記す。 1)SART-1ペプチド:SART-1抗原中にHLA-A24拘束性CTLを誘導できるペプチド分子を同定した。HLA-A26サブタイプの全てにおいてSART-1ペプチドがCTL誘導能を有することを明らかにした。また、腎癌、脳腫瘍、乳癌におけるSART-1抗原の発現を明らかにした。 2)SART-2ペプチド:SART-2遺伝子と同抗原ペプチドを解析した。100kdのSART-2抗原はERに選択的に局在し、扁平上皮癌の殆どと一部の肺癌に発現しており、一方正常組織ではその蛋白発現が認められないことより扁平上皮癌ワクチンとして臨床応用可能と判断される。 3)SART-3ペプチド:SART-3遺伝子及び同抗原ペプチドを解析した。一方、各種癌におけるSART-3抗原の発現を明らかにした。 4)SART-4(lck)ペプチド:転移性癌に高発現する癌拒絶抗原としてのSART-4(lck)分子の解析と転移癌ワクチンとしてのペプチドを同定した。本研究により食道癌に対する特異的免疫機構の分子基盤が構築され、HLA-A24陽性癌に対するペプチドを基盤とした特異的免疫療法の可能性が大きく開拓されたものと考えられる。
