悪性脳腫瘍に対する自殺遺伝子とケモカイン遺伝子を併用した電気遺伝子療法の開発

研究概要	1) 遺伝子導入条件の確立ラットグリオーマ細胞株C6の皮下腫瘍モデルを作成し、luciferase発現プラスミド及びGFP発現プラスミドを用いて、導入効率の改善をはかった。生体内電気穿孔法用の電極は6本の電極を有するneedle arrayを用いて、電極に囲まれる範囲に均一な電場が形成されるようにした。電気穿孔のパラメーターである電圧とパルスの幅を変化させた。パルス数は8で固定した。また、パルスの間隔も1secで固定した。これにより、50msec,75Vのパルスが最も有効で、単にプラスミドを腫瘍内に注入した場合よりも160倍のluciferase活性を得ることが出来た。GFPの発現分布も腫瘍内の広い範囲に確認された。電場を回転させることにより、回転しない場合の約2倍の導入効率が得られることが明らかとなった。腫瘍内に注入するプラスミドの量は50μgが滴量であった。 2) 自殺遺伝子を用いた治療実験上記の如く確立した方法を用いて、治療実験を行った。HSV-TK遺伝子を発現プラスミドpCEP4に組み込み、この実験に使用した。皮下腫瘍内にプラスミドを注入後、上記の条件で電気穿孔を行い、遺伝子を導入した。48時間後からGnciclovirを3mg/100gb.w.x2/day i.p.で投与し、治療を行った。腫瘍径を2日おきに測定し、腫瘍の増殖を測定した。コントロールとして、プラスミド注入のみにGcvを投与するもの、Gcvのみを投与するもの、電気穿孔法のみを加えるものを設けた。腫瘍の増殖は有意に抑制され、12個の腫瘍中4個の腫瘍は消失した。遺伝子導入の回数を増やすことにより、治療効果は有意に上昇した。今後は、ケモカイン遺伝子を加えて、治療の実験を行う予定である。
