悪性脳腫瘍に対する自殺遺伝子とケモカイン遺伝子を併用した電気遺伝子療法の開発

研究概要

1)自殺遺伝子を用いた治療実験
ラットおよびマウス皮下腫瘍モデルを作製し、ルシフェラーゼ発現プラスミドを用いて、生体内電気穿孔法による遺伝子導入における最適な電気的パラメーターを決定した。さらに、GFP(green fluorescent protein)発現プラスミドを導入し、腫瘍内でのGFPの発現分布を検討した。以上で決定したパラメーターを用いて、ジフテリア毒素Aフラグメント遺伝子(DT-A)、単純ヘルペスウイルス・チミジンキナーゼ(HS-tK)遺伝子を用いた治療実験を行った。双方で有意な腫瘍増殖抑制が認められたが、特にHS-tK遺伝子を導入し、48時間後からGnciclovir(3mg/100g b.w.x2/day i.p.)投与を行ったマウスでは、バイスタンダー効果も付加され50-90%という著しい腫瘍の抑制効果が得られた。12個の腫瘍中4個の腫瘍は消失した。遺伝子導入の回数を増やすことにより、治療効果は有意に上昇した。また、この方法では、プラスミドの利点として、一定の期間をあけての繰り返し遺伝子導入が可能であること、治療効果は初期治療時の腫瘍サイズに依存し、その原因として固形腫瘍内での遺伝子の発現が不均一であること、などの知見を得た。本研究により、電気遺伝子療法は実際の生体内で効果的であることが明かとなった。
2)自殺遺伝子とサイトカイン遺伝子を用いたcombination electro-gene therapy
プラスミドを用いた遺伝子療法の大きな利点の一つとして、各種のcombination gene terapyが、プラスミドを混合するのみで可能であるという点が有る。上記の自殺遺伝子を用いた治療にIL-12発現プラスミドをを併用して行うcombination electro-gene therapyの治療実験を行った。IL-12はp40およびp30をそれぞれ別のプラスミドに組み込んで使用した。従って、combination therapyではHSV-TK遺伝子の発現プラスミドを含めて3種類のプラスミドを同時に導入した。治療効果はHS-tk単独の際よりも遙かに強力な抗腫瘍効果が得られた。腫瘍が消失した個体においては、腫瘍細胞に対する全身性の免疫の成立が確認された。
