体外受精胚および凍結融解胚の生存発育能向上を目的とした研究

研究概要	卵自身の内分泌代謝活動を研究する方法を各種開発し、特に非代謝性のグルコースを用い1個の卵のグルコース取り込み能の発達解析法を確立した。卵を培養発育させ、各種パラメーターを用い体内発育卵と比較した。培養液に各種物質を添加卵の発育生存能に対する効果を検索した。さらに最近問題となっているダイオキシンを培養系に添加して低濃度でも胚発育に影響することを明らかにした。受精卵が発育するにつれて取り込み能が発達すること、体外培養によって発育した卵では取り込み能発達に遅延があり着床能も低いことを明らかにした.この発達遅延は糖輸送担体GLUTの発現が遅れることが原因であることを微量ウエスタンブロット法、RT-PCR法により蛋白、mRNAレベルで明らかにした。体外培養の条件に問題があると考え環境因子や生理活性物質の検定を行なったところ増殖因子であるepidermal growth factor(EGF)がGLUTの発現を促進しグルコース取り込みを亢進し着床能も増加させ、生理活性物質であることが明らかになった。ヒトを含めた未発育の卵はグルコースの利用性が悪く、発育途上で利用能が高まることが知られた。2-DGの取り込みを用いた動物実験では、胚発育に伴い数百倍の上昇を認めた。この変化は糖輸送担体(GLUT1)の発現と代謝酵素ヘキソキナーゼ活性によることが本研究で明らかになった。この発達は培養卵では遅延し糖取り込み能が卵発育生存能検定へ応用できることが示唆された。またエストロゲン受容体α、βともに初期胚に発現していることも確認し、内分泌撹乱物質の作用点である可能性を示した。
