体外受精胚および凍結融解胚の生存発育能向上を目的とした研究

研究概要	胚自身の内分泌代謝活動を研究する方法を各種開発し、特に非代謝性のグルコースを用い、1個の胚のグルコース取り込み能の発達解析法を確立した。胚を培養発育させ、各種パラメーターを用いて体内発育胚と比較した。培養液に各種物質を添加し胚の発育生存能に対する効果を検索した。その結果、増殖因子であるepidermal growth factor(EGF)がGLUTの発現を促進し、糖取り込み能を向上させ、その結果着床能も向上させることが明らかとなった。ヒトを含めた未発育の胚は糖の利用能が悪く、発育途上で利用能が高まることが知られた。2-deoxyglucoseを用いた動物実験では、胚発育に伴い数百倍の上昇を認めた。この変化は糖輸送担体(GLUT1)の発現量の上昇によることを微量ウェスタンブロット法、定量的RT-PCR法により蛋白、mRNAレベルで明らかにした。この発達は体外培養胚、凍結融解胚では遅延し、糖取り込み能が胚の生存発育能検定へ応用できることが示唆された。プログラムフリーザーを用いたslow法(S法)と液体窒素中に直接投入するVitrification法(V法)がマウス初期胚の生存発育能に及ぼす影響を比較検討した。V法により凍結後融解した2細胞期胚はS法凍結融解胚および新鮮胚に比べて胞胚発育率が有意に低下した。また、得られた胞胚は形態および細胞数、trophoblastの発育進展には差がなかったが、2-DGを用いて解析した糖取り込み能は新鮮胚、S法、V法の順で有意に低下し、胚移植による着床能もこれと平行して低下した。これより胚の凍結保存はその生存発育能に後々まで影響を及ぼし、特にV法はS法よりも胚に与える傷害が大きいことが示唆された。本研究で用いられた糖取り込み能は、胚の凍結融解法の改善に有用な示標になると思われた。
