歯根膜感覚を伝達する三叉神経節ニューロンの、機能と中枢投射の様態との関連の解明(免疫電顕法、電顕的定量解析法と電気生理学的手法を用いて)

研究概要	ネコを用いた実験により、速順応性と遅順応性を弁別した歯根膜求心性線維をHRPで軸索内染色した。下位三叉神経感覚核(中間核及び尾側核)内で標識された歯根膜求心性線維終末の軸索瘤を確認した後、電子顕微鏡試料を作製した。エポン包埋された切片を光学顕微鏡で観察すると、歯根膜求心性線維終末の軸索瘤は、下位三叉神経感覚核内に比較的密に局在した。遅順応性の歯根膜求心性線維の同定は困難であり、10年度で同定されたものはわずか一例であった。11年度では、遅順応性歯根膜求心性線維の試料の作製、観察、分析に重点を置く予定である。歯根膜求心性線維終末軸索瘤の染色性が良好で、比較的密に存在する標本を選び出し、電子顕微鏡にて観察した。エポン包埋された切片から歯根膜求心性線維終末軸索瘤の密に存在する部分を切り出し、ウルトクミクロトームで約70nmの連続切片を作製し、終末が作るシナプスの連続写真を撮影した。この連続写真をイメージスキャナーにてコンピュータに取り込み、解析ソフト(NIH Image)にて、軸索瘤の体積、active zoneの面積及びその二次元的形態、シナプス小胞の体積、ミトコンドリアの体積などを定量的に解析した。11年度は、免疫電顕法を用いて、歯根膜求心性線維終末が受けているシナプス前終末に、抑制性神系伝達物質と考えられているグリシン、GABAが存在しているか、検出を試みる。また、すでに明らかになっているほかのニューロンのデータと比較し、得られた研究成果を学会及び学術雑誌に発表する。
