唾液腺における水チャネルの神経性調節機構と老性動態

研究概要

各種刺激に伴うアクアポリン-5(AQP5)の細胞内移動、構造変化や機能変化の機序を明らかにし、唾液分泌におけるAQP5の役割を解明する目的で研究し、下記の成果を得た。
1)AQP5の細胞内局在
ラット耳下腺管腔膜(APM)、基底膜(BLM)、細胞内顆粒膜(ICM)をSDS-PAGEに供し、ニトロセルロース膜に転写後、抗AQP5抗体を用いてウエスタンブロッティングにて検索したところAPMとICMにおいてAQP5の局在が認められ、BLMには認められなかった。
2)刺激に伴うAQP5の細胞内移動
耳下腺切片をアセチルコリン(ACh)、エピネフリン(EPi)と加温振盪し、上述の方法で解析するとAChでは反応開始後15秒でAQP5はICMよりAPMへ移動した。ところが、EPiでは1分後に最大に達して、10分後には減少傾向を認めた。そして、この移動にはM3-ムスカリン受容体とα1-アドレナリン受容体を介する情報伝達機構が関与していた。
3)AQP5の細胞内移動と情報伝達系
M3-又はα1-作用薬によるAQP5の細胞内移動はU-73122、TMB-8、ダントロレンで抑制されたが、H-7、GF 109203Xでは抑制されなかった。従って、この移動には細胞内貯蔵部位よりCa^2が遊離され[Ca^<2+>]iが上昇して惹起されることが示唆された。
4)AQP5の細胞内移動に関与する因子
AChやEPiによるAQP5の細胞内移動はCytochalasin DやTubulozole-Cで阻害され、この移動への細胞骨格の関与が示唆された。
