研究概要

スフィンゴシン1-リン酸(S1P)受容体として3種(Edg-1、Edg-3、Edg-5)が既に報告されていたが、今回、我々は新たにオーファン受容体であったEdg-6に対してもS1Pが特異的リガンドであることを証明した。我々はこのようにS1P受容体が複数存在する理由を調べる研究プロジェクトの一貫として、これらの受容体をCHO細胞に強制発現し、受容体サブタイプとシグナル伝達系との関連を調べた。その結果、以下の知見を得た。 (1)受容体発現細胞と標識S1Pとの結合実験より、最大結合数もほぼ同程度で、いずれの受容体もS1Pとの親和性は約10〜30nMであることが判明した。 (2)ホスホリパーゼC/カルシウム系の活性化作用、アデニル酸シクラーゼ促進、抑制応答、細胞移動を指標にS1P作用を調べたところ、たとえば、ホスホリパーゼC/カルシウム系活性化はEdg-3>Edg-5>Edg-1>Edg-6発現細胞の順でS1P作用が発現したが、細胞移動能に対してはEdg-3=Edg-1であり、Edg-5、Edg-6発現細胞ではS1P作用は観察されないなど、それぞれの受容体サブタイプはやや選択的にシグナル伝達系を刺激することが判明した。
