GPI-アンカー型補体制御因子(CD59)のシグナル伝達機構:二量体形成の意義

研究概要	GPI-アンカー型蛋白質はglycosyl phosphatidylinositolを介して細胞膜に結合したユニークな脂質蛋白質群で、細胞膜上のraftと呼ばれるシグナル伝達構造に局在することが知られている.近年、これらの蛋白質が、シグナル伝達分子として機能することが明らかにされ注目されているが、そのメカニズムについては、明らかでない。申請者はGPI-アンカー型蛋白質であるCD59が細胞表面でdimerを形成していることを明らかにし、刺激によるdimer形成が、raftのdynamicな変化をひきおこし、シグナル伝達を誘導するという仮説を提唱している。本研究ではdimer形成とシグナル伝達機構の関係を明らかにすることを目的とする. 本年度は、従来の抗体刺激に代わる、ligandによるシグナル伝達測定系の開発を試みた。CD59はC9に結合し、C9分子の重合を阻害することにより補体系を抑制することから、C9分子がCD59の細胞活性化の際のligandとなりうる可能性がある。そこで、C9ペプチドの効果を「細胞内Ca濃度の上昇」を指標として測定した。その結果、ペプチド2(407-436)がJurkat細胞で、細胞内Ca^<2+>を上昇することが明らかとなった。至適濃度は50μMであった。細胞外のCa^<2+>を除くと、細胞内Ca濃度の上昇は50%程度抑制されたことから、Caは細胞外、細胞内、両者に由来すると考えられる。また、Jurkat細胞では、架橋試薬を用いてCD59とassociationしている蛋白質を架橋、可溶化後、Western blottingでassociation分子を検出したところ、CD59 dimerとともに、60-81 kDaの新たな架橋産物を検出した。今後association分子の同定を行うとともに、チロシンキナーゼの活性化、細胞増殖などへのペプチド効果についても検討する。
